DONNA of course. Sarah Rafferty is both gorgeous and hilarious. Also Rachel, although she needs to move out of the "side kick zone" pretty soon.
Olivia Wilde's Dr. Remy "Thirteen" Hadley from House. No longer a sidekick, but she was most certainly the best of the ducklings over the course of her run.
Gillian Jacob's Britta Perry on Community. Being the straight (wo)man on a sitcom is hard, but she nails it easy.

@Gabrielle I agree about Nathan from Haaven even though Haven isn't reviewed here...
I also love Cas from Supernatural...I know he is presumed dead, but he's been helping the brothers in seasons 4 and 5. Often in season 6 too
