Season 2: Doc's death, the prom
Season 3: Derek coming into Meredith's kitchen after getting out of the quarantine, and telling her he's in love with her; the scene after Cristina's wedding, when she discovers he's gone
Season 4: The clinical trial where Jeremy died and Beth decided to go through with the treatment, and when the gay soldier died and the father didn't want his boyfriend to see him...well, basically all the scenes from "The Becoming", including the speech Cristina gives the Chief after Burke wins the Harper Avery, about how underappreciated she is and how she feels like a ghost
Season 5: Meredith leaving William's execution; Callie saying she felt short, and Mark telling her to "walk tall"; Bailey telling Melinda that it was time to let Jackson go, and saying she crossed a line trying to get him a new liver; Alex breaking down and telling Meredith that that is not how he and Izzie are supposed to have a baby; Izzie and Alex's wedding

I would say definitely Season 5 finale!
One of the most shocking, moving and intense scenes I've ever seen in a TV Show... the moment when we see George in an Army uniform, OMG! It made me seat without any movement for at least 3 minutes after it finished! I was too stunned to cry the first time I saw it, but I cried everytime rewatching it.

Now that I think about it, I also cried when George's dad died. And then any time they even showed George, I started crying again. LOL, I'm pathetic!!

to Blizz - I'm with you, I was too schocked to cry the first time around when "007" was revealed to be the John Doe. I spent the last minutes of the show sceaming "OMG!!! OMG!!! OMG!!!" Since then I have cried everytime I've watched it. (5 times and counting)
